1.MECHANISM OF ACTION
This peptide competitively binds to the "a" polymerization pocket (D-domain hole) within the β- and γ-chains of fibrinogen/fibrin with higher affinity than the native fibrin α-chain knob, thereby occupying the polymerization site and sterically blocking the normal "knob-hole" interaction required for fibrin monomer assembly. By forming a stable GPRP–fibrinogen complex, it prevents further fibrin fiber formation and inhibits Factor XIIIa-mediated crosslinking of fibrin chains, while the resulting complex exhibits increased resistance to plasmin-induced proteolysis, making it a valuable modulator of the fibrinolytic cascade in mechanistic studies.
