Magnolin functions as a direct ATP-competitive inhibitor of extracellular signal-regulated kinases ERK1 and ERK2, with IC₅₀ values of approximately 87 nM and 16.5 nM, respectively. By occupying the active pocket of these kinases, it disrupts the Ras/ERKs/RSK2 signaling axis, leading to downstream inhibition of NF-κB transactivation and reduced expression of pro-inflammatory cytokines (TNF-α, PGE₂). This mechanism underlies its ability to suppress cancer cell migration, invasion, and transformation. In renal protection models, it also exhibits anti-apoptotic effects by modulating caspase-3 and Bcl-2 activity.
