1.MECHANISM OF ACTION

[Pro³⁴]-NPY exerts its biological effects through a dual mechanism, combining broad-spectrum NPY receptor activation and Phosphodiesterase inhibitor peptide activity. The Pro³⁴ substitution optimizes the peptide’s conformational stability, enhancing its binding affinity to all NPY receptor subtypes (Y1, Y2, Y4, Y5)—G-protein-coupled receptors (GPCRs) widely distributed in the central nervous system, cardiovascular tissues, and peripheral organs. Upon receptor binding, it activates Gi/o protein signaling pathways, inhibiting adenylate cyclase to reduce intracellular cyclic adenosine monophosphate (cAMP) levels and modulating calcium ion channels to regulate cellular excitability.

2.PRODUCT INDICATIONS

NPY receptor research: Investigation of Y1/Y2/Y4/Y5 receptor structure-function relationships, serving as a standard ligand for activation assays, agonist/antagonist screening, and stability validation in cross-species studies.

Neuroscience research: Study of NPY-mediated pathways in appetite regulation, anxiety, stress response, and neuronal excitability, leveraging its Phosphodiesterase inhibitor peptide property to dissect cAMP-related signaling.

Cardiovascular research: Evaluation of NPY-induced vasoconstriction and blood pressure modulation, supporting studies on cardiovascular pathologies linked to NPY signaling and phosphodiesterase activity.

3.CLINICAL EFFICACY OF PRODUCT

[Pro³⁴]-NPY is widely utilized in preclinical research, with proven efficacy in human and rat in vitro and in vivo models. As a Phosphodiesterase inhibitor peptide, it exhibits stronger inhibitory activity against calmodulin-stimulated phosphodiesterase than native NPY, providing a reliable tool for dissecting cAMP-related signaling pathways in neuronal, cardiovascular, and metabolic cells.
