HBVpol655, HBV polymerase (655 – 663)

HBVpol655, also designated as HBV Polymerase (655-663), is a synthetic 9-amino acid peptide antigen corresponding to residues 655-663 of the Hepatitis B Virus (HBV) DNA polymerase/reverse transcriptase protein. This sequence represents a well-defined, immunodominant cytotoxic T lymphocyte (CTL) epitope, known to be presented by the HLA-A*0201 major histocompatibility complex (MHC) class I molecule.

1.MECHANISM OF ACTION

This peptide functions as a specific antigenic target for CD8+ cytotoxic T lymphocytes (CTLs). During viral replication within infected hepatocytes, the HBV polymerase protein undergoes intracellular proteasomal degradation. The resulting HBVpol655 epitope is transported into the endoplasmic reticulum via TAP transporters, loaded onto HLA-A0201 molecules, and presented on the cell surface. As an exogenous reagent, the synthetic peptide can be used to pulse antigen-presenting cells (APCs), facilitating its presentation via HLA-A0201. Specific CTLs bearing complementary T-cell receptors (TCRs) recognize this peptide-MHC complex.

    2.PRODUCT INDICATIONS

  • HBV-Specific T-Cell Response Analysis: Detecting, quantifying, and functionally characterizing HBV polymerase-specific CD8+ T cells in the peripheral blood or liver of HLA-A*0201-positive individuals with acute, chronic, or resolved HBV infection using assays like ELISPOT, intracellular cytokine staining (ICS), or tetramer staining.

  • Therapeutic Vaccine Evaluation: Assessing the immunogenicity of this epitope as a candidate component in preclinical and clinical studies of peptide-based, dendritic cell-based, or nucleic acid-based therapeutic vaccines designed to treat chronic hepatitis B.

  • Immune Escape Monitoring: Investigating the impact of antiviral therapy-induced polymerase mutations (e.g., from nucleos(t)ide analogs) within or near this epitope on T-cell recognition and viral persistence, a key mechanism of immune evasion.

  • T-Cell Receptor (TCR) Studies: Isolating and characterizing natural TCRs specific for this epitope from patient samples or using it to validate the specificity of engineered TCRs for adoptive T-cell therapy (ACT) approaches.

    3.CLINICAL EFFICACY OF PRODUCT

  • Research & Diagnostic Utility: Its efficacy lies in its reliable use as a reagent to measure a specific, clinically relevant antiviral T-cell response. Robust pol655-specific CTL responses are associated with viral clearance in acute infection and better immune control in chronic infection.

  • Therapeutic Vaccine Target: This epitope is included in several investigational therapeutic vaccine platforms for chronic HBV. The goal is to stimulate or broaden these T-cell responses to achieve a functional cure. Clinical trials have utilized vaccines containing polymerase epitopes to assess safety and immunogenicity.

  • Monitoring Immune Reconstitution: In clinical research, this peptide is used to monitor the restoration of antiviral T-cell function in patients undergoing novel immunotherapies (e.g., checkpoint inhibitors, therapeutic vaccines) combined with standard antiviral drugs.

  • Pathogenesis Insight: Studying T-cell responses to this epitope helps understand the mechanisms of T-cell exhaustion and functional impairment in chronic HBV infection, guiding the development of strategies to reverse exhaustion.
