Uroguanylin, human CAS NO.154525-25-4

<p>Uroguanylin, human (CAS 154525-25-4) is a 15-amino acid endogenous peptide hormone, primarily expressed in the gastrointestinal tract and kidneys. It belongs to the guanylin peptide family and functions as a natural ligand for the guanylyl cyclase-C (GC-C) receptor. Its primary physiological role is to regulate electrolyte and fluid homeostasis by stimulating cyclic guanosine monophosphate (cGMP) production.&nbsp…

1.MECHANISM OF ACTION

Uroguanylin exerts its biological effects by binding and activating the guanylyl cyclase-C (GC-C) receptor, a transmembrane enzyme expressed predominantly on the apical membrane of intestinal and renal epithelial cells. This binding activates the receptor's intracellular catalytic domain, converting guanosine triphosphate (GTP) into the second messenger cyclic guanosine monophosphate (cGMP). Elevated intracellular cGMP then activates protein kinase G (PKG) and type II cGMP-dependent protein kinase (cGKII).

    2.PRODUCT INDICATIONS

Intestinal Secretion Studies: Investigating the regulation of chloride and fluid secretion in intestinal epithelial cell lines (e.g., T84, Caco-2) or ex vivotissue models.
GC-C Receptor Pharmacology: Serving as the native, high-affinity agonist in receptor binding assays, cGMP accumulation studies, and functional assays to characterize GC-C signaling.
Cystic Fibrosis (CF) Research: Studying alternative chloride secretory pathways that may bypass the defective CFTR in CF, as uroguanylin can activate CFTR-independent channels in some contexts.
Renal Physiology Research: Examining the role of uroguanylin in sodium homeostasis, diuresis, and natriuresis in kidney cell models.

    3.CLINICAL EFFICACY OF PRODUCT

Physiological Role: It is a key regulator of intestinal fluid secretion and renal sodium excretion. Its release postprandially is thought to link intestinal salt intake with renal salt excretion, a concept known as the "gut-renal axis."
Therapeutic Analogs: The understanding of uroguanylin's mechanism directly led to the development of stable GC-C receptor agonists:
Linaclotide (Linzess®) and Plecanatide (Trulance®) are FDA-approved drugs for chronic idiopathic constipation (CIC) and irritable bowel syndrome with constipation (IBS-C). These are structurally related, acid-stable analogs of uroguanylin/guanylin that mimic its action locally in the intestine.
Proven Clinical Efficacy: Linaclotide and plecanatide have demonstrated significant clinical efficacy in increasing bowel movement frequency, improving stool consistency, and reducing abdominal pain in large-scale clinical trials. They work by the same final pathway (GC-C/cGMP/CFTR activation) as uroguanylin.
