gp100 (639-647)

gp100 (639-647) is a synthetic 9-amino acid peptide antigen representing residues 639-647 of the human melanoma-associated antigen gp100 (PMEL17), with the sequence ITDQVPFSV. This peptide is a well-characterized, immunodominant epitope that is naturally processed and presented by the HLA-A*0201 major histocompatibility complex (MHC) class I molecule. It serves as a specific target for cytotoxic …

1.MECHANISM OF ACTION

This peptide functions as a specific antigenic determinant for CD8+ cytotoxic T lymphocytes (CTLs). Within melanoma cells, the intracellular proteasome processes the full-length gp100 protein, generating this 9-mer fragment, which is then transported into the endoplasmic reticulum, loaded onto HLA-A0201 molecules, and presented on the cell surface. As an exogenous synthetic peptide, it can be directly loaded onto the HLA-A0201 molecules of antigen-presenting cells (APCs) in vitro. The resulting peptide-MHC complex is specifically recognized by T-cell receptors (TCRs) on gp100 (639-647)-specific CTLs.

    2.PRODUCT INDICATIONS

Melanoma Antigen-Specific T-Cell Studies: Analyzing the frequency, functional avidity, and memory phenotype of gp100 (639-647)-reactive CD8+ T cells in the peripheral blood or tumor microenvironment of melanoma patients.
In VitroCTL Generation & Expansion: Stimulating and expanding antigen-specific human CTL lines or clones from HLA-A*0201-positive donor PBMCs for use in functional cytotoxicity assays and adoptive cell therapy research.
Immune Monitoring & Validation: Serving as the target antigen in standardized immune monitoring assays, including interferon-gamma ELISPOT, intracellular cytokine staining (ICS) by flow cytometry, and HLA-A*0201 tetramer staining for tracking antigen-specific T-cell responses in clinical trial samples.
Cancer Vaccine Development: Evaluating the immunogenicity and protective efficacy of this epitope in preclinical peptide-based, dendritic cell-based, or nucleic acid-based vaccine models for melanoma.

    3.CLINICAL EFFICACY OF PRODUCT

Immunogenic Validation: This epitope is one of several defined immunogenic peptides from gp100 that have been shown to consistently elicit specific CD8+ T-cell responses in HLA-A*0201+ melanoma patients and healthy donors, confirming its relevance as a tumor-associated antigen.
Clinical Trial History: The gp100 (639-647) epitope, often alongside other gp100 peptides, has been incorporated into numerous clinical trials of peptide-based vaccines for metastatic melanoma, frequently administered with immune adjuvants (e.g., Montanide ISA-51) or cytokines (e.g., IL-2, GM-CSF).
Therapeutic Modality Context: While standalone peptide vaccines have shown limited objective response rates, this epitope remains a critical target for more potent adoptive T-cell therapy (ACT). Clinical trials utilizing T-cells engineered with TCRs recognizing gp100 epitopes, including the 639-647 epitope, have demonstrated measurable tumor regression in a subset of patients with metastatic melanoma.
