gp100 (178-187)

gp100 (178-187) is a synthetic 10-amino acid peptide antigen corresponding to residues 178-187 of the human melanoma-associated antigen gp100 (PMEL17). This peptide, with the sequence VLYRYGSFSV, is a well-defined, immunogenic epitope presented by the HLA-A*0201 major histocompatibility complex (MHC) class I molecule. It is recognized by specific cytotoxic T lymphocyte…

1.MECHANISM OF ACTION

This peptide acts as a specific antigen for cytotoxic T lymphocytes (CTLs) within the framework of anti-tumor immunity. In melanoma cells, the full-length gp100 protein is processed intracellularly, and this specific 10-mer fragment is presented on the cell surface bound to the HLA-A0201 molecule. As an exogenous peptide, it can be loaded onto HLA-A0201 on professional antigen-presenting cells (APCs) in vitro. The gp100 (178-187)/HLA-A0201 complex is specifically recognized by complementary T-cell receptors on CD8+ T cells.

    2.PRODUCT INDICATIONS

  • Melanoma-Specific T-Cell Research: Investigating the frequency, phenotype, and functionality of gp100 (178-187)-reactive T cells in melanoma patients or healthy donors.

  • In VitroT-Cell Stimulation & Expansion: Generating and expanding antigen-specific human CTL lines or clones from HLA-A*0201+ donor PBMCs for use in cytotoxicity assays and functional studies.

  • Immune Monitoring: Serving as the target antigen in sensitive immune assays such as enzyme-linked immunospot (ELISPOT) for IFN-γ, intracellular cytokine staining (ICS) by flow cytometry, or HLA tetramer staining to track antigen-specific T-cell responses in clinical trial samples or preclinical models.

    3.CLINICAL EFFICACY OF PRODUCT

  • Preclinical Validation: This epitope has been validated in numerous in vitroand animal studies as capable of eliciting strong, tumor-specific CTL responses, establishing its relevance as a melanoma-associated antigen.

  • Clinical Trial Context: Alongside other gp100 epitopes (like 154-162 and 209-217), the 178-187 peptide has been incorporated into various cancer vaccine clinical trials for metastatic melanoma, often in combination with immune adjuvants or cytokines like IL-2.

  • Therapeutic Modality: While peptide vaccine trials have shown limited objective response rates as monotherapies, this epitope remains a prime target for more potent adoptive T-cell therapy (ACT). In ACT, T-cells are engineered with TCRs recognizing this epitope, leading to demonstrated tumor regression in some patients in clinical studies.
