Fibronectin Analog CAS NO.125455-58-5

<p>Fibronectin Analog (CAS: 125455-58-5), also known as Fibronectin Fragment III1-C or FN III1-C, is a recombinant or synthetic peptide fragment derived from the first type III domain of fibronectin. This 30-40 amino acid sequence contains the RGD (Arg-Gly-Asp) motif and adjacent synergy sites that mimic the cell-binding domain of native fibronectin. It exhibits high affinity for integrin receptors (particularly α5β…

1.MECHANISM OF ACTION

The Fibronectin Analog functions primarily through integrin receptor binding via its RGD motif. The RGD sequence (Arg-Gly-Asp) directly interacts with the ligand-binding pocket of integrins α5β1 and αvβ3, triggering intracellular signaling cascades. This engagement leads to focal adhesion complex formation, activation of focal adhesion kinase (FAK), and recruitment of talin and vinculin to adhesion sites. Additionally, the synergy region adjacent to RGD enhances binding affinity and specificity for α5β1 integrin through conformational changes.

2.PRODUCT INDICATIONS

Cell Culture Surface Coating: Enhancing attachment and proliferation of primary cells (endothelial cells, fibroblasts, mesenchymal stem cells) in vitro

Biomaterial Functionalization: Modifying synthetic scaffolds, hydrogels, and medical devices to promote cell integration and tissue regeneration

Integrin Signaling Studies: Investigating integrin-mediated cell adhesion, migration, and mechanotransduction pathways
Wound Healing Assays: Supporting cell migration in scratch wound and transwell migration assays
Stem Cell Differentiation: Promoting osteogenic and chondrogenic differentiation of mesenchymal stem cells on modified surfaces
Angiogenesis Models: Supporting endothelial cell tube formation and vascular network development

3.CLINICAL EFFICACY OF PRODUCT

In vitro studies demonstrate concentration-dependent enhancement of cell adhesion (2-3 fold increase compared to uncoated surfaces) and accelerated wound closure in scratch assays
Animal models of skin wound healing show improved re-epithelialization and collagen deposition when treated with RGD-containing peptide coatings
Bone regeneration studies in rodent defect models indicate enhanced bone formation and mineralization with peptide-functionalized scaffolds
Cardiovascular applications suggest improved endothelialization of vascular grafts coated with fibronectin analogs
