Fibronectin Adhesion-Promoting Peptide CAS.125720-21-0

Fibronectin Adhesion-Promoting Peptide (also known as Heparin Binding Peptide, WQPPRARI) is a synthetic 8-amino acid peptide derived from the carboxy-terminal heparin-binding domain of fibronectin. This peptide sequence (Trp-Gln-Pro-Pro-Arg-Ala-Arg-Ile) exhibits potent cell adhesion-promoting properties by interacting with cell surface heparin sulfate proteoglycans and integrin receptors. It is widely used in cell c…

1.MECHANISM OF ACTION

The peptide functions through dual mechanisms: heparin sulfate proteoglycan (HSPG) binding and integrin-mediated signaling. The PRARI core sequence within the peptide (positions 4-7) binds to cell surface HSPGs, particularly syndecans, which act as co-receptors. This binding triggers intracellular signaling cascades involving focal adhesion kinase (FAK) and Src family kinases, leading to cytoskeletal reorganization and focal adhesion formation.

2.PRODUCT INDICATIONS

Cell Culture Enhancement: Coating culture surfaces to improve attachment and proliferation of primary cells (endothelial cells, mesenchymal stem cells, fibroblasts) that are difficult to culture
Tissue Engineering Scaffolds: Functionalization of biomaterials (hydrogels, polymer scaffolds) to promote cell infiltration and tissue integration
Wound Healing Models: In vitro studies of cell migration and extracellular matrix remodeling in wound repair assays
Stem Cell Research: Promoting aggregation and spheroid formation of mesenchymal stem cells for 3D culture applications
Angiogenesis Studies: Supporting endothelial cell tube formation and vascular network development in vitro

3.CLINICAL EFFICACY OF PRODUCT

In vitro studies show concentration-dependent enhancement of endothelial cell adhesion (up to 70% increase compared to uncoated surfaces) and migration in scratch wound assays
Animal models of skin wound healing demonstrate accelerated re-epithelialization and improved granulation tissue formation when peptide-functionalized dressings are applied
Bone regeneration studies in rodent defect models show improved osteogenic differentiation of mesenchymal stem cells when cultured on peptide-modified scaffolds
Cardiovascular applications indicate enhanced endothelialization of vascular grafts coated with this peptide sequence
